Resolution of nucleotide sugars and oligosaccharides by lectin affinity chromatography.

چکیده
Bandeiraea simplicifolia I (BS I) isolectins, immobilized on Sepharose beads, specifically retarded low molecular weight ligands containing terminal cu-o-galactopyranosyl and 2acetamido-2-deoxy-a-D-galactopyranosyl residues. A BS I lectin-Sepharose column has been used to perform very efficient separations of structurally homologous sugar nucleotides and oligosaccharides. For example, UDP-glucose and UDP-galactose (and the corresponding 2-acetamido-2-deoxy derivatives) were separated by as much as three column volumes by a BS I lectin-Sepharose column. This column has also been used to resolve the alditols of lactose (P-o-Galp-(1 4 4)-D-Gk) and melibiose (a-o-Galp-(1 + 6)-~431~). to analyze the radiochemical purity of UDP-galactose and UDP-N-acetyl-D-galactosamine preparations, and to analyze the relative proportions of UDP-N-acetyl-D-glucosamine and UDP-N-acetyl-o-galactosamine in the UDP-N-acetyl-o-hexosamine pool isolated from cultured cells.

Purification and characterization of potato lectin.
Potato lectin (Solanum tuberosum agglutinin, STA), purified by affinity chromatography on tri-N-acetylchitotriose-Sepharose 6B, has Mr approximately 100,000, as estimated by gel filtration on Sephadex G-150 and is an aggregating system with a monomer Mr = 54,000, as estimated by sedimentation equilibrium analysis. Equilibrium dialysis showed that STA (dimer) has two binding sites for a specific...
متن کاملGalactose recognition by a tetrameric C-type lectin, CEL-IV, containing the EPN carbohydrate recognition motif.
CEL-IV is a C-type lectin isolated from a sea cucumber, Cucumaria echinata. This lectin is composed of four identical C-type carbohydrate-recognition domains (CRDs). X-ray crystallographic analysis of CEL-IV revealed that its tetrameric structure was stabilized by multiple interchain disulfide bonds among the subunits. Although CEL-IV has the EPN motif in its carbohydrate-binding sites, which i...
متن کاملA New Galactose-Specific Lectin from Clerodendrum infortunatum
Background: The ethno-medical significance of Clerodendrum genus raises the interest towards the characterization of its seed lectin by inexpensive and most effective technique. Objective: The focus of this study is the purification, characterization, and evaluation of the antioxidant and antiproliferative potential of a galactose-specific lectin from Cler...
متن کاملHigh glucose disrupts oligosaccharide recognition function via competitive inhibition: a potential mechanism for immune dysregulation in diabetes mellitus.
Diabetic complications include infection and cardiovascular disease. Within the immune system, host-pathogen and regulatory host-host interactions operate through binding of oligosaccharides by C-type lectin. A number of C-type lectins recognise oligosaccharides rich in mannose and fucose - sugars with similar structures to glucose. This raises the possibility that high glucose conditions in di...
متن کاملGlucosylated free oligosaccharides are biomarkers of endoplasmic- reticulum alpha-glucosidase inhibition.
The inhibition of ER (endoplasmic reticulum) alpha-glucosidases I and II by imino sugars, including NB-DNJ (N-butyl-deoxynojirimycin), causes the retention of glucose residues on N-linked oligosaccharides. Therefore, normal glycoprotein trafficking and processing through the glycosylation pathway is abrogated and glycoproteins are directed to undergo ERAD (ER-associated degradation), a conseque...
